Effect of 12-0-Tetradecanoylphorbol-13-Acetate on the Lipid Metabolism of Cultured Human Fibroblasts







Abstract
The effect of the tumor promoter 12-0-tetradecanoylphorbol-13-acetate on the lipid metabolism of cultured human fibroblasts was studied. This phorbol ester induced an increase of the incorporation of acetate into free fatty acids and neutral lipids; an increase in the amount of neutral glycerolipids; and an enhanced stimulation of phospholipid metabolism. The stimulating effect of 12-0-tetradecanoylphorbol-13-acetate on phospholipid metabolism may in part be due to an increase of the intracellular inorganic phosphate. The distribution of fatty acids was generally similar to the composition of calf serum used in the culture media and the composition of fatty acids in plasma membrane of cultured human fibroblasts was changed by 12-0-tetradecanoylphorbol:13-acetate.
These results suggest that TPA may cause a stimulating effect on the lipid metabolism, in particular, on the fatty acid composition in plasma membrane of cultured human fibroblast cells.
